TMPT Enzyme Immunoassay

A competitive micro-well immunoassay for the quantitative determination of TPMT activity in erythrocytes.
TPMT is a cytoplasmic enzyme that catalases the S-methylation of aromatic and heterocyclic sulfhydryl compounds. Variation in TPMT activity is genetic but may also be affected by some conditions. TPMT enzyme deficient individuals cannot tolerate thiopurine drugs such as 6-mercaptopurine or azathioprine, sometimes resulting in death. Thiopurine is commonly used in the treatment of leukaemia, myasthenia gravis and a variety of dermatological disorders, amongst others, so it is important to measure levels prior to treatment.
The kit is to be used on erythrocytes prepared from lysed red blood cells, and is based on the following catalysis reaction:
6-mercaptopurine (6-MP)--------------------------> 6-methylmercaptopurine (6-MMP)
One unit of TPMT is defined as a formation of 1nmol of 6-MMP per ml of packed red blood cells per 60 min incubation at 37ºC.
